Collagen and Elastin Structural Protein Autofluorescence
Symptom
Strong broad-spectrum autofluorescence in collagen and elastin-rich tissues including skin, lung, vessel walls, and fibrotic tissue. Emission persists through fixation and processing.
Common Causes
1High content of structural proteins (collagen and elastin)
2Broad excitation and emission spectra of these proteins
3Fibrotic tissue remodeling
4Background persists through standard fixation and processing
Solutions
1Favor far-red or near-IR fluorophores for target detection
2Reserve blue and green channels only for very bright or high-density targets
3Apply Sudan Black B (0.05-0.3% in 70% EtOH, ~10 min) for broad background reduction
4Test cupric sulfate (1-10 mM in 50 mM ammonium acetate buffer pH ~5, 10-30 min)
