Symptom
ChIP-seq generates high background signal across the genome with poor enrichment at expected binding sites. Signal-to-noise ratio is low, and peaks are difficult to distinguish from background.

1Antibody affinity is insufficient for immunoprecipitation conditions
2Antibody specificity is poor, binding to off-target proteins or epitopes
3Antibody has not been validated for ChIP or ChIP-seq applications
4Antibody dilution is not optimized for ChIP protocol

1Verify that the antibody is validated for ChIP or ChIP-seq (check vendor datasheet)
2Review antibody datasheet for validation data, recommended dilutions, and application-specific controls
3Test multiple antibodies against the same target to identify the best performer
4Use positive control targets with known binding sites to assess antibody performance
5Optimize antibody amount per IP reaction within the recommended range
