Incorrect Fc Blocking Timing and Sequence

Symptom
Reduced blocking efficacy with higher than expected background despite using Fc blocking reagent. Non-specific staining patterns similar to samples without Fc block.
Common Causes
  1. 1 Fc blocking reagent added after primary antibody staining instead of before
  2. 2 Insufficient incubation time for Fc block to saturate receptors before antibody addition
  3. 3 Simultaneous addition of Fc block and antibodies, preventing effective receptor saturation
  4. 4 Antibodies already bound to Fc receptors before blocking reagent can act
Solutions
  1. 1 Always add Fc blocking reagent before antibody staining step
  2. 2 Incubate cells with Fc block for recommended time (typically 5-10 minutes at 4°C or room temperature) before adding antibodies
  3. 3 Do not wash out Fc block before antibody staining; add antibodies directly to blocked cells
  4. 4 Follow workflow: wash cells → add Fc block → incubate → add antibodies (without intermediate wash)
  5. 5 Include Fc blocking step in written staining protocols to ensure consistent timing
