Common Causes
1Insufficient mixing during wash steps leaving proteins bound to beads
2Too few wash cycles performed (fewer than 3-4 washes)
3Wash buffer removed too quickly without adequate contact time
4Incomplete removal of wash buffer between steps leaving residual proteins
Solutions
1Wash thoroughly by placing lid on tube and inverting 5-10 times before centrifuging
2Perform at least 3-4 wash cycles with adequate wash buffer volume (10-20× bead volume)
3Allow 2-3 minutes contact time during each wash before centrifugation
4Remove all supernatant carefully using fine pipette tip after each wash
