Persistent high background with many non-specific protein bands in IP eluate despite following standard protocol, indicating residual unbound proteins remain on beads. Common causes: 1) Insufficient mixing during wash steps leaving proteins bound to beads 2) Too few wash cycles performed (fewer than 3-4 washes) 3) Wash buffer removed too quickly without adequate contact time 4) Incomplete removal of wash buffer between steps leaving residual proteins Solutions: 1) Wash thoroughly by placing lid on tube and inverting 5-10 times before centrifuging 2) Perform at least 3-4 wash cycles with adequate wash buffer volume (10-20× bead volume) 3) Allow 2-3 minutes contact time during each wash before centrifugation 4) Remove all supernatant carefully using fine pipette tip after each wash
