1Total lysate protein exceeds recommended range of 10-500 µg per IP reaction
2Too many cells used for lysis relative to IP scale (>10^7 cells)
3High protein concentration lysate used without appropriate dilution
4Antibody-bead binding sites saturated leading to non-specific protein retention

1Use recommended amount of 10-500 µg cell lysate per IP reaction as starting point
2Reduce number of cells used for lysis to 10^6-10^7 cells per IP
3Dilute high-concentration lysates with lysis buffer to optimal working concentration
4Optimize lysate:antibody:bead ratio through titration experiments
