Non-Specific Bands from Antibody Cross-Reactivity

Symptom
Multiple bands appear at unexpected molecular weights that do not correspond to known forms of the target protein. These bands may vary in intensity between experiments and do not correlate with expected protein expression patterns.
Common Causes
  1. 1 Antibody binds to off-target proteins with similar epitope sequences
  2. 2 Unpurified antibody preparations contain multiple immunoglobulin species
  3. 3 Polyclonal antibodies recognize multiple unrelated proteins
  4. 4 High antibody concentration increases non-specific binding
  5. 5 Insufficient blocking allows antibody to bind membrane non-specifically
Solutions
  1. 1 Use blocking peptide competition (pre-incubate antibody with immunizing peptide) to identify specific bands - only true target bands should disappear
  2. 2 Validate with knockout lysates or cells lacking endogenous target protein expression
  3. 3 Switch to affinity-purified antibodies or recombinant monoclonal antibodies for higher specificity
  4. 4 Optimize antibody dilution (try 2-5× higher dilution) to reduce non-specific binding
  5. 5 Increase blocking time (4 hours or overnight at 4°C) or use higher blocking agent concentration (5-10% milk or BSA)
