Weak Lysis Method Causing Nonspecific Bands

Symptom
Insufficient lysis strength results in incomplete protein extraction, leading to weak target signal and increased nonspecific bands from partially solubilized proteins.
Common Causes
  1. 1 Lysis method too mild for complete protein extraction
  2. 2 Insufficient detergent concentration in lysis buffer
  3. 3 Incomplete disruption of membrane-associated or nuclear proteins
Solutions
  1. 1 Use stronger lysis method (e.g., 1% SDS hot method) for more complete protein extraction
  2. 2 For nuclear proteins, prepare nuclear lysates using enrichment protocol
  3. 3 Increase detergent concentration or add stronger denaturants to improve solubilization and reduce nonspecific bands
