Incomplete Cell Fixation Due to Insufficient Incubation
Symptom
Cells show inconsistent staining patterns, continued metabolic activity, or poor storage stability when fixation incubation time is too short. Inadequate fixation may also fail to inactivate infectious samples properly.
Common Causes
1Fixation time <15 minutes for 4% PFA insufficient for complete cross-linking
2Fixation time <45 minutes for 1% PFA insufficient for full cell stabilization
3Incomplete penetration of fixative into dense cell pellets
4Low fixative concentration combined with short incubation time
5Inadequate inactivation of infectious agents (e.g., HIV) with <0.37% PFA
Solutions
1Incubate for 15-20 minutes minimum with 4% PFA at room temperature
2Incubate for 45-60 minutes minimum with 1% PFA at room temperature
3Use ≥0.37% PFA for at least recommended time when disinfecting HIV-infected samples
4Resuspend cell pellets thoroughly before adding fixative to ensure penetration
5Increase fixation time proportionally when using PFA concentrations below 1%
6Verify fixation completion before proceeding to permeabilization or storage
