Yield or Quality Issues from Incorrect Bead Drying

Symptom
DNA yield is lower than expected, or downstream applications show inhibition. Under-dried beads may carry over ethanol or contaminants, while over-dried beads result in poor DNA release during elution.
Common Causes
  1. 1 Beads dried for less than 5 minutes with tubes on magnetic rack, leaving residual ethanol
  2. 2 Beads dried for more than 5 minutes, becoming over-dried with matte, light brown appearance and visible cracks
  3. 3 Tube caps closed during drying step, preventing ethanol evaporation
  4. 4 Beads removed from magnetic rack during drying, preventing proper airflow
Solutions
  1. 1 Dry beads for exactly 5 minutes while tubes remain on magnetic rack with caps open
  2. 2 Visually inspect beads: optimally dried beads appear shiny and uniformly dark brown
  3. 3 Avoid over-drying: stop at 5 minutes even if small ethanol droplets are visible
  4. 4 If over-dried (matte appearance with cracks), reduce drying time in subsequent extractions
