Poor cfDNA Binding from Inadequate Incubation

Symptom
Cell-free DNA yield is suboptimal despite proper bead addition and reagent preparation. A significant portion of DNA may remain unbound in the supernatant during the binding step.
Common Causes
  1. 1 Sample and bead-binding mix incubation time shorter than the required 10 minutes
  2. 2 Insufficient mixing during the 10-minute binding incubation period
  3. 3 Inadequate contact between cfDNA molecules and bead surface during binding
  4. 4 Premature magnetic separation before binding equilibrium is reached
Solutions
  1. 1 Ensure full 10-minute incubation of sample with bead-binding mix to create optimal binding conditions
  2. 2 Mix gently during the incubation period if using a rotator or mixer platform
  3. 3 Avoid shortening the binding incubation time even when processing multiple samples
  4. 4 Set a timer to verify the complete 10-minute binding incubation for each sample batch
