Plate or Tube Melting and Adhering to Block

Symptom
After PCR program completion, plates or tubes are found melted or stuck to the thermal cycler block, making removal difficult and potentially damaging samples. Plastic shows signs of thermal degradation.
Common Causes
  1. 1 Suboptimal polypropylene quality unable to withstand thermal cycling temperatures
  2. 2 Rapid heating and cooling cycles in fast PCR protocols exceeding plastic thermal tolerance
  3. 3 Defective production lot with compromised material specifications
Solutions
  1. 1 Ensure PCR tubes/plate wells are made of high-grade molecular biology-grade virgin polypropylene
  2. 2 Try different production lot and contact manufacturer if issue persists for batch investigation
  3. 3 Use plate removal tool to carefully extract stuck vessels without sample damage; verify plastic specifications match fast PCR requirements
