Calcium Salt Crystal Formation in Serum-Free Media

Symptom
Insoluble calcium sulfate (CaSO4) crystals form in serum-free media preparations. Crystalline precipitates appear as visible artifacts under microscopy and may alter media composition by removing essential calcium.
Common Causes
  1. 1 CaCl2 and MgSO4 reacting directly in solution to form CaSO4 crystals
  2. 2 Improper order of component addition during media preparation
  3. 3 Autoclaving concentrated salt solutions causing precipitation
  4. 4 pH instability exacerbating calcium salt crystallization
Solutions
  1. 1 Dissolve CaCl2 in deionized water separately before adding other components
  2. 2 Add media components one at a time with mixing between additions
  3. 3 Sterilize calcium-containing solutions by filtration rather than autoclaving
  4. 4 Maintain stable pH during media preparation and storage
