Evaporation-Induced Salt Crystallization on Culture Surfaces

Symptom
Crystal precipitates form readily on culture flask and plate surfaces. Media concentration increases due to water loss, causing salt components to exceed solubility limits and crystallize on plastic surfaces.
Common Causes
  1. 1 Inadequate humidification in cell culture incubator (below 80-95% humidity)
  2. 2 Unsealed or poorly sealed cultureware allowing evaporation
  3. 3 Extended culture periods without media replacement
  4. 4 Incubator door left open or frequent opening reducing humidity
Solutions
  1. 1 Monitor and maintain incubator humidification at 80-95% relative humidity
  2. 2 Ensure flasks and plates are properly sealed to prevent desiccation
  3. 3 Use sterile water pans in incubator to maintain humidity levels
  4. 4 Minimize incubator door opening time and frequency
