Granular Precipitate on Cells

Symptom
Small granular precipitate visible microscopically on cells after adding transfection complexes
Common Causes
  1. 1 Excess EDTA present in DNA dilution buffer
  2. 2 Excess cationic lipid present during complex formation
  3. 3 EDTA concentration >0.3 mM in TE buffer
Solutions
  1. 1 Dilute DNA in water instead of TE buffer
  2. 2 If TE preferred, use EDTA concentrations <0.3 mM
  3. 3 Ensure cationic lipid concentrations do not exceed recommended amounts
  4. 4 Note: precipitate presence/absence is not indicative of transfection performance
