Uneven or Irregular Blot Appearance

Symptom
Membrane shows fingerprints, fold marks, or forceps imprints creating irregular patterns. Uneven signal distribution not related to protein bands.
Common Causes
  1. 1 Touching membrane with bare hands transfers oils and proteins
  2. 2 Folding or creasing membrane during handling
  3. 3 Forceps with sharp tips damage or mark membrane
  4. 4 Membrane drying unevenly during incubation
Solutions
  1. 1 Always wear powder-free gloves when handling membranes
  2. 2 Use blunt-end stainless steel forceps for membrane manipulation
  3. 3 Handle membrane gently; avoid folding or creasing at any step
  4. 4 Use sufficient reagent volumes to completely cover membrane during all incubations; ensure membrane remains wet
