Common Causes
1Protein aggregates in blocking reagent solution (e.g., BSA, milk powder not fully dissolved)
2Aggregates or precipitates in HRP-conjugated secondary antibody stock
3Insufficient wash steps allowing particulate matter to remain bound to membrane
4Touching membrane with bare fingers or gloves, leaving protein/oil residues
Solutions
1Filter blocking reagent solution through 0.2 µm or 0.45 µm Millex syringe filter before use; prepare fresh blocking solution
2Filter secondary antibody solution through 0.2 µm or 0.45 µm Millex syringe filter before use; centrifuge antibody stock briefly to pellet aggregates
3Increase number and duration of wash steps; use sufficient wash volumes to completely cover membrane
4Always handle membrane with clean blunt-end forceps; never touch membrane surface with fingers or gloves
