Weak or Diminished Western Blot Signal

Symptom
Expected protein bands appear faint or barely visible on film or imaging system, even after extended exposure times.
Common Causes
  1. 1 Blocking reagent has affinity for target protein, obscuring epitope accessibility
  2. 2 Antibody concentration too low or antibody inactivated by multiple freeze-thaw cycles or bacterial contamination
  3. 3 Insufficient antibody incubation time
  4. 4 Fluorescent secondary antibodies exposed to light during storage, causing photobleaching
  5. 5 Outdated or improperly stored chemiluminescent substrates with reduced sensitivity
  6. 6 Sodium azide present in buffers inhibiting HRP-conjugated antibodies
Solutions
  1. 1 Switch blocking agent (e.g., try casein or protein-free blocker instead of BSA), or reduce blocking time and concentration
  2. 2 Increase primary and secondary antibody concentrations; prepare fresh antibody aliquots
  3. 3 Extend antibody incubation time (e.g., overnight at 4°C for primary antibody)
  4. 4 Protect fluorescent antibody stock vials and aliquots from light using foil or amber tubes
  5. 5 Use fresh chemiluminescent substrate and verify expiration dates; store at recommended temperature
