Steps

1
Understand the control vs. reporter script purpose

The script evaluates autophagy-dependent responses in control and reporter lines based on the red-to-green fluorescent ratio within vacuoles. It assumes videos 1 through 4 have already been completed.

2
Open the script in RStudio

Launch RStudio and select the control_vs_reporter.r script from the project.

▶ 00:33
3
Source the demo data file

Use the Code menu, choose Source, and select the control_vs_reporter_demo file to run the analysis.

▶ 00:41
4
Review the summary graph output

Navigate to the output folder to view a summary graph showing normalized treatment ratios relative to vehicle for both control and reporter lines.

▶ 00:57
5
Examine printed statistical summaries

The script prints a full summary and a per-seedling summary, including an unpaired two-tailed Student's t-test comparing log-transformed normalized means between groups. These files support downstream statistical analysis tailored to your experimental design.
