Overview of trace fear conditioning protocol
Introduction to the hippocampal performance task with timeline overview. Describes conditioning day procedures, 24-hour context testing, and 48-hour tone-trace testing.
▶ --:--
2
Configure freeze frame software and shock parameters
Open freeze frame software, select trace fear conditioning protocol, and configure shock intensity to 0.5 mA. Set up the equipment with proper meter range and routing settings.
▶ 00:53
3
Prepare chamber and conduct conditioning trial
Clean the conditioning chamber with 10% ethanol, place mouse in chamber, enter subject details, and run the conditioning protocol with five tone-trace-shock trials.
▶ 02:08
4
Post-trial chamber cleaning and mouse removal
Remove mouse from conditioning chamber after trial completion and clean the chamber thoroughly with 10% ethanol between successive animals.
▶ 03:03
5
Perform 24-hour contextual memory test
Conduct context protocol testing 24 hours after conditioning. Place mouse in chamber for 300 seconds without tone or shock stimuli.
▶ 03:29
6
Prepare dark chamber for tone-trace test
Set up testing 48 hours after conditioning using dark chambers with black plastic covering the grid floor to create a new context. Set up infrared camera for recording.
▶ 04:20
7
Execute tone and trace protocol test
Select tone and trace protocol, place mouse in dark chamber, record animal behavior with infrared camera during the test, then remove mouse and clean chamber with water.
