Remove the culturing medium
After the 30-minute incubation period, remove the existing culturing medium from the imaging dish. This prepares the cells for the switch to live-cell imaging medium.

Add imaging medium to the dish
Add approximately 2 ml of phenol red-free emitting medium to the imaging disk. This specialized medium is required for proper live-cell imaging without fluorescence interference.

Prepare dish for transport to microscope
Prepare the imaging dish for transport by placing it in a protective holder with a p100 petri dish as the base and a p60 dish as the top. Cover the top with electrical tape to block ambient light during transport.

Begin live-cell imaging
The sample is now ready for imaging on the microscope. Proceed to the imaging station and begin live-cell imaging.
