Set up all materials on ice and perform a mid-line incision from posterior to anterior along the scalp to expose the skull.

Cut and open the cranium
Make careful cuts through the lateral cranial structures from neck to nose, and perform additional cuts toward the occipital region to gain full access to the brain.

Extract the intact brain
Carefully remove the whole brain from the skull using a small spatula.

Dissect and clean brain tissue
Use fine dissection forceps to remove the cerebellum, brainstem, and white matter to isolate clean brain cortices.

Wash brain tissue in PBS
Perform a quick rinse of the dissected brain tissue in non-sterile PBS to remove debris.

Homogenize brain tissue completely
Use a potter with pestle and glass tube to homogenize the fresh brain tissue into a uniform suspension.
