Launch CFX Manager and access protocol setup

Open the CFX Manager software and navigate to the protocol setup section by clicking on 'User Defined' and then 'Edit Selected' to begin making changes to your experimental protocol.

Review default qPCR protocol structure

Examine the default protocol template which includes an initial heating step followed by 40 repeats of denaturation and annealing cycles. Understand the basic structure before making any modifications.

Edit temperature and time parameters

Click on specific text fields within each step to directly edit temperature or time values. Type your desired parameters and press Enter to confirm each change.

Configure additional step options

Access the 'Step Options' menu for the current step to specify additional parameters beyond basic temperature and time settings.

Insert new steps before or after current step

Select whether to add a new step before or after the current step using the dropdown menu. Highlight the appropriate step and click 'Insert Step' to add it.

Delete unnecessary steps and adjust sample volume

Remove unwanted steps by clicking 'Delete Step'. Adjust the sample volume by modifying the value in the 'Sample Volume' box as needed for your experiment.

Review runtime estimate and save protocol

Check the estimated runtime displayed in the software to verify your protocol duration. Save your finalized protocol to complete the setup process.
