How to connect HiTrap Fibro PrismA unit to an ÄKTA system
Protocol
Difficulty
intermediate
Steps
1
Review HiTrap Fibro PrismA unit specifications
Understand that HiTrap Fibro PrismA units are ready-to-use systems designed for rapid monoclonal antibody capture and compatible with ÄKTA chromatography systems. Note that no drop-to-drop connection is required as air will be automatically flushed during high-flow operation.

2
Disconnect flow restrictor and prepare unit
Manually disconnect the flow restrictor from the unit to minimize back pressure, or use the pH valve if available. This step is essential before connecting to the ÄKTA system.
▶ 00:40
3
Wash out storage solution with low flow
Run the system at low flow rate to wash out the 20% ethanol storage solution from the unit. Air removal is automatic during this washing process.

4
Increase flow rate stepwise with leak checks
Gradually increase the flow rate using a stepwise approach, checking for leakage at each increment before proceeding to higher flow rates. Stop and investigate any leaks before continuing.
▶ 01:20
5
Establish recommended operating flow rate
Set the system to the recommended operating flow rate of 16 milliliters per minute for HiTrap Fibro PrismA operation.
▶ 01:33
6
Record baseline pressure reference values
Check and record the pre-column and delta-column pressure values as reference measurements before starting your chromatography run.
