What you'll learn
✓Apply agar disk diffusion test to measure antimicrobial material activity
✓Perform ISO 22196:2007 contact method for surface antimicrobial assessment
✓Prepare and validate microbial suspensions for testing protocols
✓Compare two complementary antimicrobial characterization methods
Protocol
We present a protocol for the antimicrobial characterization of advanced materials. Here, the antimicrobial activity on material surfaces is measured by two methods that complement each other: one is based on the agar disk diffusion test, and the other is a standard procedure based on the ISO 22196:2007 norm.

Steps
1
Prepare advanced material samples for testing
Cut and sterilize material samples according to protocol specifications. Ensure uniform dimensions and proper handling to avoid contamination prior to antimicrobial testing.
▶ 00:46
2
Perform agar disk diffusion test on materials
Place material samples on inoculated agar plates and incubate to measure inhibition zones. This diffusion-based method qualitatively assesses antimicrobial activity around material surfaces.
▶ 01:16
3
Verify microbial suspension concentration and purity
Prepare and standardize microbial suspensions by measuring optical density and confirming culture purity. Ensure suspensions meet required specifications before contact testing.
▶ 03:27
4
Measure antimicrobial activity using ISO 22196:2007 contact method
Inoculate material surfaces with standardized microbial suspension and incubate under controlled conditions. Quantify surviving bacteria to assess antimicrobial efficacy per ISO standard protocol.
▶ 04:40
5
Compare diffusion and contact method results
Analyze and interpret data from both complementary methods to validate antimicrobial characterization. Discuss how each method provides distinct information about material performance.
