DOI: 10.3791/1712-v
What you'll learn
✓Set up crystallization trials for membrane proteins in lipidic mesophases
✓Prepare and load protein-lipid mixtures into dispensing syringes
✓Dispense crystallization samples onto plates for structure determination
Protocol
Herein is described the procedure implemented in the Caffrey Membrane Structural and Functional Biology Group to set up manually crystallization trials of membrane proteins in lipidic mesophases.
Review the theoretical background and rationale for using lipidic mesophases in membrane protein crystallization trials. This foundation supports understanding of subsequent preparation and handling steps.

2
Gather required materials and reagents
Assemble all necessary equipment, materials, and reagents for the crystallization setup, including crystallization plates, syringes, lipids, precipitants, and protein samples.
▶ 02:48
3
Prepare the crystallization plate
Set up and organize the crystallization plate with appropriate wells and protective measures to prevent contamination and evaporation during sample dispensing.
▶ 03:48
4
Prepare lipid and protein mixture
Mix the membrane protein with lipid in the correct stoichiometry and concentration, ensuring homogeneous dispersion in the lipidic mesophase before loading into syringes.
▶ 05:12
5
Load protein-lipid mixture into syringe
Transfer the prepared lipid-protein mixture into the dispensing syringe using precise techniques to avoid air bubbles and ensure accurate dosing of crystallization samples.
▶ 13:53
6
Dispense samples onto crystallization plates
Carefully dispense the lipid-protein mixture onto the prepared crystallization plates following the established protocol to create conditions suitable for crystal formation.
▶ 17:57
7
Observe and monitor crystal formation
Document initial crystal appearance and growth patterns following dispensing, establishing baseline conditions for subsequent structure determination experiments.
