Protocol

The reversible addition of palmitate to proteins is an important regulator of intracellular protein trafficking. This is of particular interest in neurons where many synaptic proteins are palmitoylated. We utilize a simple biochemical method to detect palmitoylated proteins in cultured neurons, which can be adapted for multiple cell types and tissues.

Steps

Extract target protein from cultured hippocampal neurons

Lyse primary cultured neurons and prepare cell lysate for immunoprecipitation. Remove detergent-insoluble material and measure total protein concentration for downstream normalization.

Perform acyl-biotin exchange with hydroxylamine cleavage

Treat immunoprecipitated protein complexes with hydroxylamine to cleave palmitoyl groups, then conjugate exposed cysteines to biotin-HPDP for detection.

Elute biotin-conjugated target protein and run SDS-PAGE

Collect biotin-labeled immunoprecipitate, separate by SDS-PAGE, and prepare samples for Western blotting on nitrocellulose or PVDF membrane.

Detect palmitoylation via Western blotting with streptavidin

Probe membrane with streptavidin-HRP and target-specific antibodies to visualize both palmitoylated and total protein levels.

Analyze and interpret palmitoylation detection results

Compare streptavidin signal intensity between treated and control samples to quantify relative palmitoylation of synaptic proteins.
