Biopharma Insights Neural stem cells (NSCs) refer to cells which can self-renew and differentiate into the three neural lineages. Here, we describe a protocol to determine NSC frequency in a given cell population using neurosphere formation and differentiation under clonal conditions.

Steps

Culture clonal sample neurospheres
Establish and maintain neural stem cell neurospheres under clonal conditions using limiting dilution to ensure single-cell origin. Cells are cultured in appropriate medium to promote self-renewal.

Prepare neurosphere-conditioned medium
Collect and process medium conditioned by neurosphere cultures to obtain growth factors and signaling molecules that support NSC proliferation and differentiation in subsequent assays.

Transfer sample neurospheres to chambered coverslip
Carefully plate individual or clonal neurospheres onto a 50-well-chambered coverslip for controlled differentiation and microscopic analysis of single clonal populations.

Differentiate sample neurospheres
Remove growth factor support and culture neurospheres under conditions promoting differentiation into the three neural lineages (neurons, astrocytes, oligodendrocytes).

Fix differentiated neurospheres
Chemically preserve differentiated neurosphere samples using appropriate fixation to enable immunofluorescence staining and morphological analysis.

Enumerate NSC frequency from clonal data
Quantify neural lineage markers in fixed clonal neurospheres and calculate the frequency of neural stem cells in the original population based on differentiation patterns.
