DOI: 10.3791/54847-v
What you'll learn
  • Culture and passage cerebral vascular endothelial cells using standardized techniques
  • Measure mitochondrial bioenergetic function in CVE cells using functional assays
  • Interpret representative data on CVE bioenergetic profiles and metabolic capacity
Protocol

Endothelial cell mitochondria are critical to maintain blood-brain-barrier integrity. We introduce a protocol to measure bioenergetic function in cerebral vascular endothelial cells.

Difficulty

Total time
~3–5 days (including cell culture, passage, and mitochondrial assessment)
Model organism
Cerebral vascular endothelial cells (CVE)
Biosafety

Steps

1
Culture and passage cerebral vascular endothelial cells

Establish and maintain CVE cell cultures from brain tissue, including routine passage and cell viability assessment. This step ensures healthy, proliferative cells for downstream bioenergetic analysis.

2
Assess mitochondrial functional bioenergetic parameters

Perform mitochondrial stress tests and bioenergetic assays on CVE cells to measure oxygen consumption rates, ATP production, and metabolic capacity. These measurements reveal the functional health of endothelial cell mitochondria.

3
Analyze and interpret CVE bioenergetic function data

Review representative bioenergetic profiles and data outputs from CVE cells, including key metabolic parameters and comparative analysis. This step demonstrates how to evaluate and contextualize results within the blood-brain-barrier integrity framework.
