✓Perform intraperitoneal injection in adult zebrafish using microsyringe equipment
✓Anesthetize zebrafish safely with cold water for surgical procedures
✓Prepare and inject experimental substances into the peritoneal cavity
✓Monitor post-injection recovery and validate injection success
Protocol
We demonstrate intraperitoneal injection into adult zebrafish. We use a 10 μl NanoFil microsyringe controlled by a Micro4 controller and UltraMicroPump III. This demonstration includes the use of cold water as an anesthetic.
Difficulty
intermediate
Total time
~15-20 min per fish (including anesthesia, injection, and recovery)
Model organism
Zebrafish (Danio rerio)
Biosafety
BSL-1
Steps
1
Prepare microsyringe and injection equipment
Set up the 10 μl NanoFil microsyringe with Micro4 controller and UltraMicroPump III. Calibrate equipment and prepare injection solution prior to anesthesia.
▶ 01:19
2
Anesthetize zebrafish with cold water
Place adult zebrafish in cold water to achieve anesthesia. Monitor loss of response to stimuli before proceeding with injection.
▶ 05:34
3
Perform intraperitoneal injection procedure
Insert the microsyringe into the peritoneal cavity at the appropriate angle and depth. Deliver the injection volume slowly and withdraw the needle carefully.
▶ 05:34
4
Monitor recovery and post-injection assessment
Return anesthetized zebrafish to normal water conditions. Observe recovery of movement and behavioral responses. Assess injection success through physiological measurements.
