Protocol
This protocol is used to isolate single atrial cardiomyocytes from the adult mouse heart using a chunk digestion approach. This approach is used to isolate right or left atrial myocytes that can be used to characterize atrial myocyte electrophysiology in patch-clamp studies.

Steps

Dissect mouse atrial appendages
Extract and isolate the right and/or left atrial appendages from adult mouse heart tissue following euthanasia and cardiac exposure.

Isolate single atrial myocytes via chunk digestion
Apply enzymatic digestion and mechanical dissociation to atrial tissue chunks to obtain individual viable cardiomyocytes for downstream analysis.

Perform whole-cell or single-channel patch-clamp recordings on isolated atrial myocytes to characterize electrical properties and ion channel function.
