This protocol describes the measurement of isometric contraction in an isolated smooth muscle preparation, using an isolated tissue bath system and computer-based data acquisition.

Steps

Prepare and calibrate the tissue bath system

Set up the isolated organ bath with proper temperature control, aeration, and computer-based force transducer calibration. Ensure all tubing, electrodes, and data acquisition are functioning.

Dissect and prepare aortic ring segments

Extract aorta tissue and carefully cut into ring segments (typically 2–4 mm), removing excess connective tissue while preserving the smooth muscle layer.

Mount aortic rings in the tension apparatus

Position each ring segment onto the apparatus hooks, set baseline tension, and allow equilibration in oxygenated physiological buffer at 37 °C for optimal contractile response.

Apply initial challenge and perform dose–response protocol

Induce smooth muscle contraction with agonist (e.g., phenylephrine) in incremental doses, recording isometric tension changes to generate a dose–response curve.

Determine EC₅₀ and analyze contractile parameters

Calculate half-maximal effective concentration (EC₅₀) and other pharmacological parameters from the tension measurements using curve-fitting software.
