DOI: 10.3791/50757-v
What you'll learn
✓Extract and isolate lipids from bat integumentary tissues using solvent extraction
✓Separate triacylglyceride lipid classes via preparative thin-layer chromatography
✓Profile and identify TAG molecular species using MALDI-TOF mass spectrometry
✓Interpret TAG composition data to characterize species-specific lipid profiles
Protocol
Biopharma Insights Mammalian integument contains solvent-extractable lipids that can provide chemical compositions characteristic of individual species. This paper presents a routine method for separating broad lipid classes isolated from integumentary tissues using thin layer chromatography and determining the triacylglyceride profile by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.

Steps
1
Extract and prepare bat integumentary lipids
Harvest integumentary tissue from bat specimens and perform solvent extraction to isolate total lipids. This yields the crude lipid fraction for downstream separation.

2
Separate lipid classes by preparative thin-layer chromatography
Apply crude lipid extracts to preparative TLC plates and develop using appropriate mobile phase to resolve broad lipid classes. Visualize bands and recover triacylglyceride fraction.
▶ 02:33
3
Analyze TAG molecular species by MALDI-TOF MS
Prepare purified TAG fraction for MALDI-TOF mass spectrometry analysis. Acquire and interpret mass spectra to determine specific triacylglyceride molecular composition and profile.
▶ 05:34
4
Interpret triacylglyceride profiles of bat tissues
Examine resultant TAG profiles from Eastern Red Bat integumentary tissues and identify species-characteristic lipid molecular signatures for comparative biochemical analysis.
