Protocols for Testing the Toxicity of Novel Insecticidal Chemistries to Mosquitoes

DOI: 10.3791/57768-v
What you'll learn
  • Execute larval contact assays to assess insecticide toxicity in immature mosquitoes
  • Perform adult topical and blood-feeding assays for adulticide evaluation
  • Conduct dose-response studies to determine toxicity thresholds and potency
Protocol

Protocols are described for assessing the toxicity of chemistries to immature and adult mosquitoes for development as new classes of larvicides, adulticides and endectocides. The protocols enable high-throughput testing of multiple chemistries at single-point dose and subsequent evaluation via dose response assay to determine toxicity on contact or via ingestion.

Difficulty
intermediate
Total time
~3–5 days (including dose-response evaluation across multiple concentrations)
Model organism
Mosquitoes (Aedes, Anopheles, or Culex species)
Biosafety
BSL-1

Steps

1
Perform larval contact toxicity assay

Expose immature mosquito larvae to test chemistries via direct contact in controlled aquatic conditions. Assess mortality and behavioral endpoints to evaluate larvicidal potency.

2
Conduct adult topical insecticide assay

Apply test chemistries topically to adult mosquito cuticle and quantify mortality and knockdown effects. Enables assessment of contact-based adulticide efficacy.

▶ 02:56
3
Execute adult blood-feeding toxicity assay

Deliver test chemistries via blood meal to assess oral/ingestion-based toxicity in blood-fed adult mosquitoes. Evaluates endectocide potential and systemic uptake.

▶ 05:16
4
Analyze dose-response data for potency determination

Evaluate toxicity results across chemistries and dose ranges to establish IC50, LD50, and relative potency rankings. Validates assay reproducibility with reference compounds.
