What you'll learn
✓Perform microslide diffusion assay for qualitative antimicrobial enzyme screening
✓Label bacterial substrates with Remazol Brilliant Blue R dye
✓Execute dye-release assay for quantitative enzymatic activity measurement
✓Interpret and characterize novel protein antimicrobial candidates
Protocol
Biopharma Insights Here, we present protocols to rapidly screen and characterize enzymes for antimicrobial activity. The microslide diffusion assay and the dye-release assay utilize target bacterial substrates for qualitative and quantitative enzymatic activity evaluation.
Difficulty
intermediate
Total time
~4–6 hours per assay set (includes bacterial culture preparation, substrate labeling, and assay incubation)
Biosafety

Steps
1
Perform qualitative microslide diffusion assay
Set up a microslide diffusion assay using target bacterial substrates to qualitatively detect enzymatic antimicrobial activity. Spot enzyme samples and observe diffusion-based inhibition zones.

2
Label substrate with Remazol Brilliant Blue R dye
Prepare bacterial substrates by labeling with Remazol Brilliant Blue R dye to enable colorimetric detection in subsequent quantitative assays.
▶ 03:37
3
Execute quantitative dye-release assay
Perform the dye-release assay using labeled bacterial substrates to quantitatively measure enzymatic activity. Quantify enzyme-mediated dye release spectrophotometrically.
▶ 05:38
4
Interpret results and characterize antimicrobials
Analyze qualitative and quantitative assay data to detect and characterize novel protein antimicrobial candidates, comparing enzymatic activity profiles.
