Steps
1
Isolate and prepare mouse skin tissue
Remove dorsal skin from mouse and prepare tissue for cell isolation. Includes cleaning, trimming, and initial dissociation steps.

Isolate primary dermal and epidermal cells
Separate dermis from epidermis and enzymatically dissociate each layer into single-cell suspensions for culture and genetic modification.

Graft modified dermal and epidermal cells
Recombine lentivirally transduced dermal and epidermal cells and implant the construct onto recipient mice to initiate hair follicle regeneration.

Perform histological analysis of hair follicles
Harvest grafted skin, process tissue sections, and conduct microscopic evaluation to assess hair follicle growth, morphology, and epithelial-mesenchymal signaling.
