Retinal Detachment Model in Rodents by Subretinal Injection of Sodium Hyaluronate

DOI: 10.3791/50660-v
What you'll learn
  • ✓ Perform subretinal injection of sodium hyaluronate
  • ✓ Create reproducible retinal detachments in rodents
  • ✓ Avoid subretinal hemorrhage during surgery
  • ✓ Monitor retinal detachment over time
Protocol

Biopharma Insights Creating experimental retinal detachments with a reproducible and sustained height of detachment, and without subretinal hemorrhage, is important for studying the pathophysiology of photoreceptor cell loss in retinal disease and evaluating potential therapeutic interventions. Here, we report such a method in detail.

Difficulty
advanced
Total time
~30-45 min/animal plus recovery
Model organism
Rodent (e.g., mouse or rat)
Biosafety

Steps

1
Prepare and anesthetize rodent for surgery

Anesthetize the rodent and position it under the microscope for subretinal injection.

▶ 02:17
2
Inject sodium hyaluronate subretinally

Perform subretinal injection of sodium hyaluronate to induce a controlled retinal detachment.

▶ 04:42
3
Assess retinal detachment time course

Observe and document the progression of retinal detachment over time.

▶ 05:16
