Use of a Hanging Weight System for Coronary Artery Occlusion in Mice

DOI: 10.3791/2526-v
What you'll learn
  • ✓ Prepare the mouse for surgical coronary occlusion
  • ✓ Expose the heart via thoracotomy
  • ✓ Apply the hanging-weight system to occlude artery
  • ✓ Confirm ischemic zone by color change
Protocol

Biopharma Insights A murine model for myocardial ischemia and ischemic preconditioning is an important tool study cardioprotective mechanisms in vivo. Here, we report an easy applicable in situ model for cardiac IP using a hanging-weight system for coronary artery occlusion.

Steps

1
Prepare mouse for surgery

Anesthetize and position the mouse, then perform endotracheal intubation and expose the surgical site.

2
Expose and occlude coronary artery

Open the chest to expose the heart, then use the hanging-weight system to occlude the coronary artery and induce ischemia.

▶ 04:33
3
Conclude and confirm occlusion

Verify successful occlusion by observing tissue blanching or ECG changes, then proceed with reperfusion or further steps.
