Xenotransplantation of Human Stem Cells into the Chicken Embryo
DOI: 10.3791/2071-v
What you'll learn
✓Perform surgical window creation in chicken embryos for CNS access
✓Execute hindbrain lesion induction and stem cell transplantation techniques
✓Assess human stem cell proliferation and differentiation in vivo
Protocol
In this paper we present a method for transplanting human stem cells into various regions of the central nervous system of the chicken embryo. This provides an in vivo model for assessing the proliferation and differentiation of various types of human stem cells in embryonic tissue environments.
Difficulty
advanced
Total time
~4–6 hours per embryo (including incubation and surgical procedures)
Model organism
Chicken embryo (Gallus gallus domesticus)
Biosafety
BSL-1
Steps
1
Introduce xenotransplantation methods and objectives
Overview of human stem cell transplantation into chicken CNS and rationale for using embryonic tissue environments to study proliferation and differentiation.

2
Create surgical access window in embryo
Perform surgical windowing technique to expose the central nervous system of the chicken embryo for subsequent transplantation and lesion procedures.

3
Induce hindbrain lesion and transplant stem cells
Create a targeted hindbrain lesion and transplant human stem cells into the lesioned region to enable assessment of cell behavior in damaged tissue.
