Steps

Amplify DNA fragment with PCR

Use polymerase and primers complementary to the DNA sequence of interest to amplify the target fragment exponentially through repeated PCR cycles. The resulting amplified DNA will have either a single-A overhang (if Taq polymerase is used) or blunt ends (if high-fidelity polymerase like Q5 is used).

Join amplified DNA to vector

Insert the amplified DNA fragments into an appropriate vector using either DNA ligase or activated vectors with covalently-bound effector enzymes. The fragment can be inserted non-directionally depending on the vector type, such as in TA cloning.

Select for successful insertion

Use suicide vectors containing toxic genes that ensure only competent cells with successful target gene insertion survive, since insertion disrupts expression of the toxic gene.

Transform recombinant plasmid into E. coli

Introduce the recombinant plasmid into competent E. coli cells using standard transformation procedures.

Plate cells on antibiotic-containing agar

Spread the transformed bacteria onto agar plates containing the appropriate antibiotic for selection of successfully transformed cells.

Screen colonies for target gene

Analyze the resulting colonies to identify those containing the gene or fragment of interest using appropriate screening methods.
