Steps

Unpack and set up the base unit

Familiarize yourself with the Trans-Blot Turbo system components including the base unit with integrated power supply, LCD screen, two transfer cassette bays, blot roller, instruction manual, and transfer packs. Plug in the power cord to the rear of the unit and turn on the power switch to prepare the instrument for use.

Open cassette and prepare membrane stack

Open a transfer cassette by turning the knob counterclockwise until you feel a click. Peel open the turbo transfer pack foil and identify the two ion reservoir stacks. Place the bottom stack containing the membrane in the cassette base with the membrane facing upward, then gently roll with the blot roller to remove trapped air.

Layer gel and top stack into cassette

Lay the gel directly on top of the membrane without equilibrating. Place the top ion reservoir stack on the gel and gently roll again to remove any bubbles between the layers. Place the cassette cover on top of the sandwich and press down firmly.

Lock cassette and insert into instrument

Turn the cassette knob clockwise until you feel a click to secure the transfer sandwich. Slide the locked cassette into one of the two bays (A or B) on the Trans-Blot Turbo base unit, which are independently controlled for simultaneous or sequential transfers.

Select protocol and initiate transfer

Access the turbo protocols using the LCD screen and keypad to select the appropriate protocol based on your gel type and size (e.g., 7-minute rapid transfer or 3-minute TGX protocol). Start the run and monitor the countdown timer and real-time voltage/amperage conditions displayed on the screen.

Remove cassette and extract blot

Wait for the beep and 'run complete' message on the LCD screen indicating transfer is finished. Remove the cassette from the instrument bay and open it by turning the knob counterclockwise. Carefully lift off the top stack to reveal the transferred blot on the membrane.

Clean cassette and customize protocols

Rinse the cassette with water and wipe down with a paper towel for easy cleanup between runs. Use the LCD screen and keypad to access pre-installed protocols or program and save custom transfer conditions by adjusting voltage, current, and time parameters for your specific experimental needs.
