Remove gel from package and prepare
Remove the Mini-PROTEAN TGX Stain-Free gel from its package and peel off the green strip from the bottom of the gel.

Install gel into electrode assembly
Secure the gel into the electrode assembly inside the tank, positioning the comb side of the gel inward toward the assembly. If running only one gel, insert a buffer dam into the opposite side.

Remove the comb from gel wells
Remove the comb by firmly pulling straight up and out of the wells, then gently straighten any well walls that may have collapsed during comb removal.

Fill buffer chambers with running buffer
Fill the inner buffer chamber completely with running buffer, then fill the outer buffer chamber to the appropriate line on the tank.

Clean wells to remove acrylamide debris
Wash out each of the wells with running buffer to remove residual acrylamide debris, which will minimize band distortion during electrophoresis.

Load samples into gel wells
Load your sample into the wells, being careful not to puncture the well bottom.

Connect leads and start electrophoresis run
Connect the electrical leads to begin the run, and ensure that bubbles are coming from both the anode and cathode, signifying that the run has started successfully.
