How to use a Pipette and a Burette

Steps

1
Distinguish pipette and burette equipment

Learn to identify the two pieces of glassware: a pipette is pipe-shaped with fixed volume and a fine line marking, while a burette has increments up to 50 ml. Understand the three buttons on the pipette filler: A (aspirate), S (suction), and E (eject).

2
Attach filler and draw liquid into pipette

Attach the pipette filler to the pipette, press the A button while deflating the bulb, then place the pipette tip into the liquid container. Press the S button to draw liquid into the pipette until it reaches the designated line, re-inflating and deflating the bulb as needed.

3
Transfer and eject liquid from pipette

Move the filled pipette to the receiving flask and press the E button to eject all liquid. Remove the pipette filler and gently tap the pipette against the flask bottom to dislodge any remaining drops.

4
Understand burette construction and operation

Learn the burette components: read the meniscus line to determine liquid volume, use the tap at the bottom to dispense liquid dropwise, and adjust the screw to tighten or loosen the tap. Practice opening the tap slowly to achieve accurate, controlled dispensing.

5
Secure burette and pipette vertically

Mount both the burette and pipette on a retort stand ensuring they are perpendicular to the table top. Step back and observe the setup from a distance to confirm both instruments are upright, not slanted.

6
Fill burette and remove air bubbles

Place a filter funnel in the top of the burette and pour solution through it, slightly lifting the funnel to allow air flow. Use the burette tap to release excess solution until the meniscus reaches the 0 ml marking, then open the tap to remove any air bubbles trapped in the tip.
