How to use a rotovap (rotary evaporator)

Protocol
Difficulty
intermediate

Steps

1
Prepare water bath and cooler

Turn on the water bath and set it to 40°C, and set the cooler to 10°C (or 1°C for low boiling solvents like ether). Ensure both systems reach the desired temperature before starting evaporation.

2
Attach adapter and secure flask

Select a glass adapter and connect it to the rotovap using a thick green ring without grease. Position the water bath by turning the metal latch until it locks firmly into place.

▶ 00:59
3
Start rotation and apply vacuum

Turn on the machine and set the flask to slow rotation. Open the tap to allow ambient air pressure, then gradually close the tap while reducing pressure and monitoring the flask for solvent evaporation into the adapter.

▶ 02:28
4
Monitor solvent distillation progress

Continue reducing pressure as solvent distills over into the cooled adapter. After most solvent has been collected, stop the process and discard the remaining solvent from the adapter.

▶ 05:30
5
Remove residual solvent under high vacuum

Reduce pressure further to approximately 50-60 milli bars to evaporate remaining solvent. Connect an oil pump to achieve final drying, and collect any additional solvent that escapes into the safety bottle.

▶ 06:55
6
Configure inert gas protection if needed

Optionally attach a nitrogen balloon to the tap for air-sensitive products, ensuring proper positioning of the tap selector to deliver nitrogen instead of air upon completion.

▶ 08:20
7
Remove product and shut down equipment

Use the button to lower the water bath away from the flask. Turn off the pump and rotation, then remove the dried product from the rotovap.

▶ 09:11
8
Clean up and complete shutdown

Turn off the main power button, water bath, and cooler. Check all connections for residual grease or solvent, empty the safety bottle, and allow components to dry completely.
