Power on and log into ChemiDoc system

Press the green power button on the ChemiDoc MP instrument. When the home screen appears, tap to access the login menu and enter your username credentials or create a new user account.

Prepare imaging chamber and clean tray

Open the imager drawer and pull out the transilluminator. Clean the UV tray with DI water and a Kimwipe to ensure optimal imaging performance.

Load membrane and configure imaging parameters

Apply substrate to the membrane and center it on the UV tray. Close the drawer and select chemiluminescence as the application under blots. Adjust image size and zoom as needed for your region of interest.

Set exposure and capture chemiluminescent image

Configure exposure settings using either automatic band detection or manual exposure time entry. Optionally use signal accumulation mode to find ideal exposure. Tap the camera button to capture the image, which automatically saves to the gallery.

Capture colorimetric marker and merge images

Capture a colorimetric image of the molecular weight marker to visualize band positions. Select your preferred chemiluminescence exposure and tap merge to combine the images for complete visualization.

Analyze band intensities and adjust appearance

Tap individual bands to view their relative fluorescence units and percent saturation values for quantification. Use the transform icon to adjust image appearance for visualization without affecting actual data.

Export and save image data to file

Tap the send/save icon and select your desired file format (including Image Lab format for further analysis). Choose destination as USB drive or network and export with any custom notes or metadata.
