High Non-Specific Background Signal in Competitive ELISA
Symptom
Elevated background signal is detected across wells, interfering with specific signal measurement. Background noise reduces assay sensitivity and may mask low-concentration analyte detection.
Common Causes
1Detector conjugate binding non-specifically to the microplate surface
2Excessive concentration of detector conjugate antibody
3Inadequate blocking buffer composition or insufficient blocking incubation
4Insufficient washing cycles or inadequate wash duration
Solutions
1Run control wells with primary antibody and sample omitted to identify non-specific conjugate binding
2Titrate and decrease the concentration of detector conjugate antibody
3Optimize blocking by testing alternative blocking buffers or extending blocking time/temperature
4Increase the number of wash cycles and/or extend wash duration per cycle
