High CV from Poorly Maintained Plate Reader

Symptom
High coefficient of variation (>15%) across the plate with no obvious pattern. Signal intensity may drift over time or show unexpected variability between identical samples.
Common Causes
  1. 1 Plate reader optics (filters, lenses) are dirty or contaminated
  2. 2 Instrument not calibrated according to manufacturer schedule
  3. 3 Light source intensity has degraded over time
  4. 4 Temperature control in reader chamber is unstable
  5. 5 Reader software or firmware requires updating
Solutions
  1. 1 Verify that the instrument has been maintained and calibrated according to manufacturer recommendations
  2. 2 Clean optical components using manufacturer-approved cleaning solutions and procedures
  3. 3 Perform calibration using standard reference plates or filters provided by manufacturer
  4. 4 Check maintenance logs and schedule service if instrument has not been serviced recently
  5. 5 Update software/firmware to the latest version recommended by manufacturer
