Symptom
No staining is detected when targeting membrane proteins in IHC. Signal is absent despite antibody validation, suggesting structural damage to membrane epitopes.
Common Causes
1Permeabilization with strong detergents (e.g. Triton X) damaged delicate membrane protein structures
2Cell membranes disrupted by excessive detergent exposure
3Membrane epitopes destroyed during harsh permeabilization procedures
Solutions
1Use less stringent detergent (e.g. Tween 20 instead of Triton X)
2Remove permeabilizing agent completely from buffers
3Optimize permeabilization conditions specifically for membrane proteins
4Test protocol without permeabilization step for surface membrane proteins
