Loss of membrane protein staining after permeabilization

Symptom
No staining is detected when targeting membrane proteins in IHC. Signal is absent despite antibody validation, suggesting structural damage to membrane epitopes.
Common Causes
  1. 1 Permeabilization with strong detergents (e.g. Triton X) damaged delicate membrane protein structures
  2. 2 Cell membranes disrupted by excessive detergent exposure
  3. 3 Membrane epitopes destroyed during harsh permeabilization procedures
Solutions
  1. 1 Use less stringent detergent (e.g. Tween 20 instead of Triton X)
  2. 2 Remove permeabilizing agent completely from buffers
  3. 3 Optimize permeabilization conditions specifically for membrane proteins
  4. 4 Test protocol without permeabilization step for surface membrane proteins
