Weak or Absent Signal Due to Insufficient Exposure
Symptom
Western blot film shows very faint bands or no visible bands after initial exposure. The membrane may contain protein and antibody binding has occurred, but the chemiluminescent or colorimetric signal is not captured adequately on film.
Common Causes
1Film exposure time too short for the amount of target protein present
2Low abundance target protein requiring extended detection time
3Weak chemiluminescent signal that decays before adequate exposure
4Film sensitivity insufficient for the signal intensity
Solutions
1Expose the film for longer duration (try incremental exposures: 1 min, 5 min, 15 min, 30 min)
2Use high-sensitivity film or switch to CCD/CMOS imaging systems
3Apply fresh chemiluminescent substrate immediately before exposure
4Optimize ECL substrate choice (use enhanced or femtogram-sensitivity substrates for low-abundance targets)
