Weak or Suboptimal IHC Staining Intensity

Symptom
Staining signal is present but significantly weaker than expected, with poor contrast between positive cells and background, making interpretation difficult.
Common Causes
  1. 1 Incorrect primary antibody dilution or use of non-recommended diluent buffer
  2. 2 Suboptimal antigen retrieval method (water bath instead of microwave; wrong buffer formulation)
  3. 3 Primary antibody incubation time too short or temperature not optimized (not overnight at 4°C)
  4. 4 Using biotin-based detection instead of more sensitive polymer-based detection system
  5. 5 Inadequate deparaffinization leaving paraffin residue that blocks antibody access
Solutions
  1. 1 Follow CST product datasheet for recommended antibody dilution and specific diluent (SignalStain® Antibody Diluent vs. TBST/5% NGS varies by product)
  2. 2 Use microwave oven for antigen retrieval; for enhanced signals, test pressure cooker method
  3. 3 Incubate primary antibody overnight at 4°C according to validated CST protocol
  4. 4 Switch to polymer-based detection (SignalStain® Boost #8114 or #8125) instead of biotin-based or directly conjugated HRP secondaries
  5. 5 Repeat deparaffinization using fresh xylene with new tissue sections
