Poor Duplicate Reproducibility

Symptom
Duplicate wells showing inconsistent results with high variation between replicates
Common Causes
  1. 1 Insufficient washing
  2. 2 Automatic plate washer ports clogged or obstructed
  3. 3 Uneven plate coating due to procedural error or poor plate quality
  4. 4 Reused plate sealers
  5. 5 No plate sealers used
  6. 6 Contaminated buffers
Solutions
  1. 1 Follow washing procedures from ELISA Development Guide page 4
  2. 2 If using automatic plate washer, check all ports are clean and free of obstructions
  3. 3 Add 30-second soak step and rotate plate halfway through wash
  4. 4 Dilute in PBS without additional protein
  5. 5 Check coating and blocking volumes, times and method of reagent addition
  6. 6 Use proper ELISA plates (not tissue culture plates)
  7. 7 Use fresh plate sealer for each step
  8. 8 Make fresh buffers
